Development of kunitz trypsin inhibitor (KTI)-free soybean is crucial for soy-food industry as the heat inactivation employed to inactivate the anti-nutritional factor in regular soybean incurs extra cost and affects protein solubility. In the presented work, a null allele of KTI from PI542044 was introgressed into cultivar ‘JS97-52’ (recurrent parent) through marker assisted backcrossing. Foreground selection in BC1F2, BC2F2 and BC3F2 was carried out using the null allele-specific marker in tandem with SSR marker Satt228, tightly linked with a trypsin inhibitor Ti locus. Background selection in null allele-carrying plants through 106 polymorphic SSR markers across the genome led to the identification of 9 KTI-free lines exhibiting 98.6% average recurrent parent genome content (RPGC) after three backcrosses, which otherwise had required 5–6 backcrosses through conventional method. Introgressed lines (ILs) were free from KTI and yielded at par with recurrent parent. Reduction of 68.8–83.5% in trypsin inhibitor content (TIC) in ILs compared to the recurrent parent (‘JS97-52’) was attributed to the elimination of KTI.  相似文献

高色素基因hp1和hp2能显著提高番茄果实的类胡萝卜素总量,对于以提高番茄红素含量为目标的品质育种具有重要应用价值。以这2个基因的突变位点为目标,成功地筛选出了对hp1和hp2分别具有高度特异性的2对引物。由于这样的分子标记实际上就是功能基因本身局部片断的扩增结果,所以能非常可靠地直接鉴别该功能基因,F2群体中分子标记的分离比例与2对基因的独立分配比例完全一致。以这两对引物所产生的分子标记为依据,已经合成了含有hp1和hp2的双突变体。  相似文献

甘蓝型油菜SRAP、SSR、AFLP和TRAP标记遗传图谱构建   总被引:27,自引:0,他引:27  
以黄籽GH06为母本、黑籽P174为父本杂交得到的第6代重组自交系188个株系为作图群体,通过SRAP、SSR、AFLP和TRAP四种分子标记对该群体进行遗传连锁分析,构建了一张包含20个连锁群、300个标记位点的甘蓝型油菜分子遗传图谱(LOD≥3.0),包括202个SRAP标记、65个SSR标记、23个AFLP标记和10个TRAP标记。图谱总长度1273.7cM,标记间平均距离为4.25cM。连锁群上的标记数在4~56个之间,连锁群长度变动在37.1~109.2cM之间,群内平均图距在1.80~14.20cM之间。LG1包含的标记最多,有56个;标记最少的连锁群(LG9、LG18、LG20)只有4个。LG13的平均图距最大,为14.20cM;LG6的平均图距最小,仅为1.80cM。在整个图谱上,存在图距大于20cM的空隙6个。本研究首次将SRAP及TRAP标记用于甘蓝型油菜遗传图谱的构建,结果表明,SRAP及TRAP标记在甘蓝型油菜遗传图谱的构建上是一种良好的标记系统。  相似文献

The sexual propagation of genebank accessions by hand pollination in the greenhouse is a very expensive and time-consuming routine task. The self-incompatibility of most diploid potato species allows their multiplication in isolated field plots, which is coupled with the advantage of the presence of natural pollinators and maintaining a high level of heterozygosity. The aim of this study was to determine the isolation distance required for minimizing pollen flow between neighbouring multiplication plots. The Solanum phureja clone IVP48 was used as the pollen donor since it possesses a dominant seed marker (‘embryo spot’). The centrally located pollen donor was surrounded by S. phureja recipient plots at distances of 10,20,40, and 80 m, respectively. The average pollen contamination ranged from 0.2% in the plots 80 m from the pollen donor to 5.1% in those 10 m away. In this study, procedures are suggested which will help to reduce cross-pollination between accessions.  相似文献

Random amplified polymorphic DNA (RAPD) analysis has proven to be an effective procedure for molecular marker applications in plant breeding, although non-specific amplification may limit its utility in some species. The objective of this study was to determine the effectiveness of restriction-endonuclease digestion of template DNA for elimination of non-specific amplification and generation of heritable RAPD markers. Restriction endonucleases digested wheat DNA to completion in amplification buffer, suggesting that the restriction endonuclease can be added directly to the reaction mixture prior to amplification. A 1-h 37°C step was programmed into the thermal cycler for restriction-endonuclease digestion which was followed immediately by amplification. Non-specific amplification was reduced and DNA marker patterns were altered in digested samples when compared to undigested samples. Genetic segregation of two polymorphic markers tested in F5 inbred progeny fit expected 1:1 ratios. These results suggest that heritable DNA markers may be generated with reduction in non-specific amplification when restriction-endonuclease digestion of template DNA is conducted as part of the RAPD procedure.  相似文献

Summary The inheritance and linkage relationships of a leaf morphology gene of Matthiola incana were investigated. The allele for sinuate leaf shape, c, was found to be recessive to the allele for normal entire leaf, C. The c allele was tightly linked to the recessive allele for double flowering, s. The recombination frequency between the two loci was close to zero. The mode of inheritance of the C gene was in accordance with the hypothesis that a pollen lethal gene is responsible for the constant 1:1 segregation ratio of double-flowered (= male sterile) to single-flowered (= fertile) plants in most M. incana breeding lines. The sinuate leaf allele seemed to reduce the frequency and delay the flowering of double-flowered plants. The importance of the C gene as a double flowering marker in the cultivation and the breeding of M. incana is discussed.Contribution from the Agricultural Research Organization, The Volcani Center, Bet Dagan, Israel. 利用ISSR标记研究鹅观草属种质资源的遗传多样性   总被引:5,自引:0,他引:5  
为进一步了解鹅观草属种质资源的遗传背景和拓宽牧草育种的遗传基础,科学指导鹅观草属种质资源的收集、评价和利用,利用ISSR(Inter simple sequence repeat marker)标记对鹅观草属20个种,6个变种,共60份材料进行了遗传多样性检测。结果发现,被测材料间ISSR标记的多态性较高,在20个引物中,有16个引物可扩增出清晰且重复性好的DNA片段,共产生125条DNA片段,其中104条具有多态性,多态性比率为83.20%;每个引物可扩增出6~10条DNA片段,平均7.81条,材料间遗传相似系数GS变幅为0.188~0.879,平均值为0.375;聚类分析表明,在遗传相似系数为0.441的水平上,60份材料可以聚为5类,属于同种、同组、同系的不同材料首先聚在一起,然后再与其他种的材料聚在一起,此外,材料的聚类还表现出一定的地域性规律。  相似文献

In the high amylose starch mutant ‘Glacier AC38’, a single recessive gene designated amo1 is responsible for an amylose content of up to 45%. A rapid technique was established in order to evaluate the amylose/amylopectin ratio in half kernels. To localize this gene, crosses with multiple marker lines and trisormes were conducted. In addition, RFLP markers were used to determine their mapping distance to amo1. Two markers are located 2 cM and 7 cM, respectively, from amo1 on chromosome 5S (1HS). The relationship between the wx and amo1 genes was also examined and the role of the amo1 gene in starch synthesis is discussed.  相似文献

The random amplified polymorphic DNA (RAPD) and microsatellite techniques were used to evaluate the genetic diversity among 18 soybean genotypes selected for a breeding programme to increase the protein content of varieties adapted for central European growing conditions. Out of 33 random primers used in RAPD reactions, only 12 showed polymorphism useful for characterization of these genotypes. In contrast, all 12 microsatellite primer pairs used in this study detected polymorphism with 2–6 alleles per locus. Similarity measures and cluster analysis were made using RAPD and simple sequence repeat (SSR) data, separately and together. The resulting dendrograms were compared with each other and with the available pedigree information as a control. The dendrogram derived from RAPD data showed some divergence from the pedigree information available for the lines. The dendrograms based on SSR data and SSR data combined with RAPD gave very good agreement with pedigree information. It can be concluded that the combined use of a limited number of RAPD and SSR markers is a useful and reliable means of evaluating genetic relationships of genotypes in the absence of pedigree data.  相似文献
